A comparative study of the determination of Metoclopramide.HCL in pharmaceutical formulations by Flow injection Spectrophotometric and Reverse phase-High performance liquid chromatography RP-HPLC methods

Abstract

This study involves development of a new flow injection spectrophotometric
method and a reverse phase-high performance liquid chromatography RP-HPLC
method for the determination of Metoclopramide.HCl in aqueous solution and in
pharmaceutical preparations.
FIA method was based on the oxidative coupling reaction of
Metoclopramide.HCl with phenothiazine in the presence of ferric nitrate in neutral
medium to form a green water soluble dye that is stable and has a maximum
absorbance at λmax 600 nm. A linear calibration graph was in the range of 2.5-100
μg.ml-1, with a correlation coefficient of 0.9998, detection limit of 1.2 μg.ml-1 and a relative standard deviation of 0.62%. The nature of the coloured produced dye was determined and found to be 1:1. The method was applied successfully for the determination of Metoclopramide.HCl in pharmaceutical preparations (mean recovery
99.85-101.00%).
In HPLC method, the drug was analyzed using RP-HPLC method with a Supelco BDS-C18 -DS (25 cm x 46 mm i.d) analytical column (5 μm particle size) and isocratic elution with a mobile phase containing 50% acetonitrile in 0.05 M KH2PO4
buffer (pH 5.5), at a flow rate of 1 ml.min-1, 20 μL sample loop, and the UV detector was set at λmax 273 nm. Calibration graph was in the range of 0.05–0.75
μg.ml-1 with a correlation coefficient of 0.9998, and a relative standard deviation of 2.37%. The method was applied successfully for the determination of Metoclopramide.HCl in pharmaceutical preparations with a recovery of 98.78% -
99.35%.
